Rapamune (Sirolimus) Uses

This medicine is an immunosuppressant. It is indicated to prevent organ rejection after a kidney transplant. It may also be used to treat certain other conditions if determined by your doctor to be indicated.

How Rapamune (Sirolimus) Works

This medication works by weakening your body's immune system. This helps your body to accept the transplanted organ.

How To Take Rapamune (Sirolimus)

You can take this medication either with or without food. However you should choose one way (with or without food) and always take it that way so the amount of drug absorbed stays constant.

It is very important to take this medication at the same time each day. This keeps the levels of drug in your bloodstream and body at a more constant level. This increases the efficacy of the drug while lowering the risk of Rapamune Side Effects.

If you are also taking cyclosporine, take this medicine 4 hours after your cyclosporine dose unless directed otherwise by your doctor.

Avoid consuming grapefruit and grapefruit juice while taking this medication. Grapefruit can alter the metabolism of drugs, resukting in increased risk of Rapamune Side Effects.

Possible Rapamune Side Effects (Sirolimus)

Diarrhea, joint pain, shaking, acne or trouble sleeping as well as other Rapamune side effects have been reported.

Notify your doctor if you experience any of these serious Rapamune side effects, including:

  • muscle pain and/or cramps,
  • bone pain,
  • increased thirst and/or hunger,
  • frequent urination,
  • vision changes,
  • hearing problems (examples: hearing loss, ringing in the ears),
  • unusual tiredness and/or weakness,
  • fast or slow or irregular heartbeat,
  • easy bruising or bleeding,
  • mental and/or mood changes,
  • swelling ankles and/or feet,
  • severe headache,
  • dizzyness,
  • stomach and/or abdominal pain.

Notify your doctor immediately if you experience any of these very Rapamune serious side effects:

  • change in the amount of urine produced,
  • frothy urine,
  • chest pain,
  • shortness of breath,
  • pain or swelling or redness of arms or legs,
  • dark urine,
  • yellowing eyes and/or skin,
  • persistent nausea and/or vomiting.

Although serious allergic reaction to this medication is unlikely, seek immediate medical attention if it occurs. Symptoms of serious allergic reaction include rash, itching, swelling, dizziness or trouble breathing.

Rapamune (Sirolimus) Warnings and Precautions

This drug is an immunosuppressant and lowers the body's ability to fight infections and diseases. An increased risk of infection and certain types of cancer may occur. To reduce these risks, your doctor will prescribe the lowest possible effective dose. This lowest dose also reduces your risk of developing serious side effects.

Notify your doctor immediately if you develop any of the following:

  • unusual growths or lumps,
  • swollen glands,
  • unusual skin changes,
  • change in the appearance or size of moles,
  • night sweats,
  • unexplained weight loss
  • signs of an infection such as fever, persistent cough or sore throat.

Rapamune can also cause cholesterol or triglycerides to increase. Have your cholesterol checked periodically. You may need cholesterol drug treatment to control this increase.

Serious allergic reactions to this drug are rare. However you should seek immediate medical attention if you experience rash, itching, swelling, trouble breathing or severe dizzyness.

Rapamune (Sirolimus) Drug Interactions

The list of drugs that interact with Rapamune (sirolimus) is extensive. Tell your doctor all of the medications you take. You should always have your medication list with you and share that list with your doctor and pharmacist.

The following drugs are known to interact with Rapamune: St.John's Wort, metoclopramide, amphoteracin B, tacrolimus, temsirolimus, gentamicin, ganciclovir, phenytoin, carbamazepine, cisplatin, rifampin, rifabutin, nelfinavir, ritonavir, nefazodone, danazol, bromocriptine, cimetidine, erythromycin, diltiazem, verapamil, nifedipine, ketoconazole, itraconazole, cisapride, cyclosporine, mycophenolate, basilimaxib.

This list is not complete. Many more drugs interact with Rapamune. Consult your doctor or pharmacist for further or more detailed questions.

Missed Dose

If you miss a dose, take your next dose as soon as you remember. If it is time for your next dose, skip the missed dose and go back to your regular schedule. Do not take double doses.

Storage

Keep this medication in the container it came in, tightly closed. Keep out of reach of children. Store at room temperature (between 68 and 77 degrees F or between 20 and 25 degrees C.) Store away from excess heat and moisture (not in the bathroom). Store this medicine out of direct sunlight. Throw out medication that is outdated or no longer needed.

Pregnancy/Nursing

This medication should not be used during pregnancy. If you suspect that you may be pregnant, talk to your doctor. It is not known whether this medication is excreted in the breast milk. It is advised not to breast-feed while taking this medication.
